Iscove's modified Dulbecco's medium/Ham's F-12 1:1, chemically defined lipid concentrate, penicillin/streptomycin, monothioglycerol (450 μM), apo-transferrin (15 μg/ml) and crystallization-purified bovine serum albumin (5 mg/ml, Sigma). The medium was supplemented with insulin (1 μg/ml) and SB431542 (10 μM, Tocris) on day 0 (the starting day of differentiation culture). |
