	MCF-7 cells were transfected with RNA duplex of stealth siRNA targeted for the human ObR mRNA sequence (Ambion, ID:s224009), with a control siRNA (Invitrogen ID:45-2001) that does not match with any human mRNA, used as a control for non-sequence-specific effects to a final concentration of 50 nM using Lipofectamine 2000 as recommended by the manufacturer. After 5 h the transfection medium was changed with serum-free medium and then the cells were exposed to treatments.
