	MITF siRNA was used to downregulate MITF expression. Cells were seeded in a 6 well plate (1 × 105 cells/well) and allowed to adhere at 37 °C for 24 h. The following day, cells were transfected with 40 nM of siMITF or siRNA-A scrambled control (Santa Cruz Biotechnology, USA) using Lipofectamine 2000 (Invitrogen, USA). 150 μg/ml GpS treatment was started after transfection. After 48 h GpS treatment, cells were harvested and intracellular melanin content was assessed.
