Protocol tips
- Cells were fixed with 4% paraformaldehyde for 20 min.

- Permeabilized with PBS containing 0.1% Triton X-100 and 0.1% glycine for 2 min on ice
Downstream tips
- When Nonspecific labelling is present, reduce concentration of TdT.

- When high background is present reduce concentration of labelling mix by 50%

Publication protocol

Sk-mel-2 cells mounted on glass slides were fixed with 4% paraformaldehyde for 20 min, and permeabilized with PBS containing 0.1% Triton X-100 and 0.1% glycine for 2 min on ice. The TUNEL staining was performed using the In Situ Cell Death Detection Kit, Fluorescein (Roche Diagnostics, Germany). Cells were co-stained with 4′6′-diamidino-2-phenylindole (DAPI) to visualize nuclei. The images were taken using a FSX100 all-in-one microscope (Olympus Corporation, Japan).

Are 10 week old samples in formalin still usable?

I have left my tumor samples in formalin for an extended period of time (around 10 weeks). Do you think I will be able to use them for TUNEL assay and get results? Thank you for your help in advance
