We used the Live/Dead Cell Imaging Kit (Molecular Probes, Life Technologies Corporation) to evaluate cell viability in our models. In the Live/Dead assay, the viable cells are stained green, while the dead cells are stained red35. The cells were loaded into the microfluidic devices, as described previously, and after one day, we used phosphate buffered saline (PBS, HyClone) to wash the culture chambers in the microfluidic chip for 1–3 min. Then, the cells were incubated with the Live/Dead Cell Imaging Kit for 15–30 min at 37 °C. Next, we used PBS again to wash out the reagent for 3–5 min and observed the culture chambers under a fluorescent microscope. Finally, cell viability was measured by assessing the percentage of green fluorescent cells in all of the cells.
