The purified S-HDAg gene and vector pPICZαA (TFS, V19520) were digested with XbaI and KpnI (NEB, R0145S and R0142S, respectively) and purified by QIAquick PCR Purification Kit (Qiagen, 28106) following the manufacturer’s instructions. The digested HDAg gene was ligated into the linearized vector pPICZαA using T4 DNA ligase (NEB, M0202S). The reaction was performed in a 20 µL volume including 2 µL 10× Rapid Ligation Buffer, 8 µL DNA (~100 ng), 1 µL 5 U/µL T4 DNA ligase and incubated at 22 °C for 2 h.
