Upstream tips
This vector includes a strong, thermo-inducible Pr promoter [31] and was co-operating with CI repressor. To improve the success rate of plasmid construction, a specific CI repressor-mutant B. subtilis strain BUSY 9797 was employed, which totally represses the expression of inserted genes.

The copy number of pGETS118 vector in B. subtilis tends to be close to one but can be increased by 1 mM isopropyl 1-thio-β-d-galactoside (IPTG). The resulting plasmids were re-purified and transformed into the expression host B. subtilis RM125.
