The plasmid, pUC8CVX-RsaAΔ0–222, was then electroporated (Gilchrist & Smit, 1991 ▸) into the expression strain C. crescentus JS1014 (Ford et al., 2007 ▸). JS1014 is a protein-expression strain that has had its genomic copy of rsaA and a surface-associated metallo­protease, sapA, knocked out; SapA has previously been found to degrade modified versions of RsaA (Ford et al., 2007 ▸). In addition, all major surface polysaccharides are eliminated by knockout of the manB gene, which prevented any possibility of RsaA surface attachment and minimized cell buoyancy.
