The protein expression of Notch signaling pathway related genes (Notch1, Hes1) in samples was detected by immunohistochemical method. IHC staining was performed using formalin-fixed paraffin-embedded blocks as previously described [11]. The following primary antibodies were used: anti-Notch1, Santa Cruz, 1 : 100 dilution; anti-Hes1, Abcam, 1 : 200 dilution. Known positive controls using normal skin tissue were included for each run, and negative controls were done by omitting the primary antibodies. Two independent observers assessed immunoreactivity using a three-grade system, where 0 denoted negative staining; 1 denoted minimal and variable staining; 2 denoted obvious and intense staining. Sections with grade 2 were considered positive staining.
