Publication protocol

Cryopreserved PDX 7482 tumor pieces were revived and surgically implanted in the mammary fat pad of a NOD/SCID female mouse for expansion for 5 weeks. To assess effects of knockdown on mammosphere formation potential, 5 × 103 cells from PDX 7482 (obtained from a tumor piece which had been expanded in a NOD/SCID mouse, harvested, collagenase treated, strained, red blood cell lysed, washed, and counted) were seeded in complete MammoCult media (Stemcell Technologies) in technical triplicate replicates in 24-well ultra-low-adherence plates (Corning).
