Protocol tips

Upstream tips
Use early passages (1–5) of primary tumor tissues from PDX models
Protocol tips
Seed PDX cells at a density of 500 cells/well
Use of 96-well ultralow attachment plates
Count spheres 5-7 days later
Downstream tips
Collect spheres for other procedures, such as RNA isolation

Publication protocol

For mammosphere assays, PDX cells were seeded on 96-well ultralow attachment plates (Corning) at a density of 500 cells/well in StemXVivo Serum-Free Tumorsphere Media (R&D, CCM012) in the presence of the indicated peptides. Images were acquired by using a phase contrast microscope (Olympus Microsystems), and spheres were counted 5-7 days later.
