"CD34+ HPs of the THC2 patient were isolated from bone marrowaspirate with monocyte-separating medium (TBD) followed by anti-
CD34+ magnetic microbead positive selection using EasySep™ Human
CD34 Positive Selection Kit II (STEMCELL). The CD34+ HPs were cultured
in StemPro™-34 medium (STEMCELL) supplemented with
StemSpan™ megakaryocyte expansion supplement (100 × ) (STEMCELL)
contains a combination of recombinant human cytokines (SCF, IL-
6, IL-9 and TPO) for transduction. The SHAMUi001-A line was generated
from the exponential phase of the growing cells. Briefly, the
CD34+ HPs were reprogrammed by the integration-free CytoTune-iPS
2.0 Sendai Reprogramming Kit (Invitrogen) which utilizes Sendai virus
particles of the four Yamanaka factors (Lieu et al., 2013; Takahashi
et al., 2007). The transduced cells were cultured on a matrigel (Cellapy)
with StemPro™-34 medium (excluding cytokines) for 3 days. Then, they
were transferred to matrigel-coated culture dishes and cultured with
the Essential 8 (E8) medium (Cellapy) to conclude the transitioning.
After 4 weeks, individual iPSC colonies were manual picked up and
expanded with E8 medium. All cells used in this study were incubated
in a humidified incubator with 5% CO2 at 37 °C."
