DNA Microarray Assay
From each group we selected four mice, who had a stratum corneum water content close to the average level. Total RNA samples from individuals were subjected to DNA microarray analysis as described previously (49). In brief, using a GeneChip 3'IVT Express Kit (Affymetrix, Santa Clara, CA), we synthesized cDNA from 200 or 300 ng purified total RNA, and biotinylated aRNA was transcribed with T7 RNA polymerase. The aRNA was fragmented and hybridized to an Affymetrix GeneChip Mouse Genome 430 2.0 Array (Affymetrix), which contained probes for >39,000 mouse genes. After hybridization at 45°C for 16 h, the array was washed and stained with phycoerythrin. Fluorescence signals were scanned with the Affymetrix GeneChip System (Affymetrix). Affymetrix GeneChip Command Console software (Affymetrix) was used to reduce array images to the intensity of each probe.
