Global DNA methylation assay
The mammary tumors and mammary tissues were collected and placed in RNAlater (Ambion) prior to freezing and then disrupted into small pieces. DNA was purified by the standard method using the GenElute Mammalian Genomic DNA Miniprep Kit (Sigma–Aldrich). Concentration was quantified using a NanoDrop 2000c (Thermo Fisher Scientific) and global levels of cytosine methylation in DNA samples were measured using the Imprint Methylated DNA Quantification Kit (Sigma–Aldrich). Briefly, DNA samples (100 ng per well) were incubated with capture and detection antibodies, and then the absorbance was measured at 450 nm with a Spectra-Max Microplate Reader (Molecular Devices).
