Publication protocol

Cells stabilized in RNAprotect were centrifuged. The resulting pellet was lysed and homogenized in a buffer containing denaturing guanidine–thiocyanate, inactivating RNases to prevent RNA degradation during processing. The lysate was then applied to a gDNA eliminator column. To facilitate appropriate binding conditions for RNA, ethanol was added to the flow-through. Finally, the sample was added to an RNeasy spin column, where total RNA binds to the membrane and contaminants are efficiently washed away. Although the gDNA eliminator column in the RNeasy Protect Cell Mini Kit removes the vast majority of DNA, trace amounts may remain, depending on the amount of the sample loaded.
