Biosensor preparation.Recombinant HAs were diluted to 40 to 100 μg/ml in 1× kinetics buffer (PBS containing 0.02% Tween 20, 0.005% sodium azide, and 100 μg/ml bovine serum albumin, purchased from Fortebio as 10× stock) and were coupled either online (during the assay) or offline (prior to running the assay) to streptavidin-coated biosensors. To achieve this, biosensors were precoupled to penta-His biotin conjugate (Qiagen, Valencia, CA) or biotin nitrilotriacetic acid (BNTA) (Biotium, Hayward, CA), according to instructions provided by Fortebio.
