Template DNA preparation and multiplex PCR assay
The genomic DNA from reference strains and clinical isolates of different species was extracted by using Gentra Puregene Yeast DNA extraction kit (Qiagen, Hilden Germany) according to kit instructions or by the rapid method using Chelex-100. A loop full of yeast colony grown on Sabouraud dextrose agar plate was suspended in 1 ml of sterile water in a microcentrifuge tube containing 50 mg Chelex-100 (Sigma-Aldrich Co., St. Louis, MO, USA), the contents were heated at 95°C for 20 min and then centrifuged. The supernatant was transferred to a new tube and typically 2 μl was used for mPCR.
