Kit Aa employed mechanical disruption of cells with bead beating and DNA binding to paramagnetic beads. Briefly, 1 ml of phosphate buffered saline was added to 300 mg of feces; the mixture was vortexed and then centrifuged. The supernatant was then added to a tube containing zirconia beads and a lysis-binding solution. Following bead-beating disruption for 15 min using a vortexg with a multitube adapterh at maximum speed, the lysate was clarified by centrifugation and added to a plate along with isopropanol, magnetic nucleic acid–binding beads, and binding-lysis enhancer. Binding, washing, and elution of the DNA were performed on an automated systemi yielding DNA in a 90-µl elution volume.
