Bisulfite sequencing and combined bisulfite restriction analysis (COBRA)
500 ng of genomic DNA from each mouse liver tissue was treated with bisulfite using EZ DNA Methylation-Gold Kit (Zymo Research, Orange, CA, USA) followed by PCR. The primers used were listed in S1 Table. For bisulfite sequencing, the PCR products of three control or TCE treated samples were pooled and cloned into TA-cloning vector (Tiangen), and individual clones were sequenced using an automatic sequencer (ABI 3730XL). For COBRA, the PCR products were digested with the corresponding restriction enzymes and analyzed on 2% agarose gel as described earlier [16].
