One of the major challenges is to make a specific construct that has no responses other than those related to the signaling pathway of interest. This can be achieved by selecting highly specific reporter constructs containing only defined responsive elements and a minimal promoter linked to reporter enzymes such as luciferase

Upstream tips
Seed 1 × 10^5 cells/well
Protocol tips
Add 2 ml of 1X Fixing Buffer and incubate 5–10 minutes at room temperature.

Add 2 ml of the X-Gal Staining Mix to the dish and incubate the dish at 37°C for 1 hr to overnight
Protocol tips
Add 2 mL of 1X Fixing Solution. Incubate at room temperature for 5 minutes.

Wash cells and add Cell
Staining Working Solution.

Incubate the cells at 37ºC protected from light for 1 hr to overnight.
